43kDa Dystrophin-associated glycoprotein; (43-DAG, DG, b-DAG, b -DG, DAG1) β-dystroglycan is a component of the dystrophin glycoprotein complex (DGC), a membrane-spanning complex that links the interior cytoskeleton to the extracellular matrix in muscle and other tissues (Winder 2001). It is a transmembrane protein (single TM domain) of 34.9kDa molecular weight, and pI 8.85, which displays an apparent molecular weight of 43kDa by SDS-PAGE due to asparagine-linked glycosylation and a high proportion of proline residues in the cytoplasmic domain. The dystroglycan complex is a subcomplex within the DGC comprising alpha and beta-subunits. Dystroglycan is of importance in cell signalling, cell polarity, cell adhesion and movement. β-dystroglycan binds to dystrophin (in muscle cells) and utrophin (in non-muscle cells), but dissociates from these proteins when phosphorylated on Tyr-892 allowing its association with SH2 containing proteins such as Src and other cytolinker proteins such as plectin. This antibody recognises Tyr-892 phosphorylated forms of β-dystroglycan (Tyr-892 in human, Tyr-890 in mouse)
Tested applications: Western blot (1:500 dilution), IHC Microscopy (1:50 dilution), Immunoprecipitation (1:10 dilution) in RIPA buffer. Species recognition: recognises β-dystroglycan in all mammalian species, when phosphorylated on Tyr-892 (or equivalent).
