Description Mesenchymal stem cells are multipotent stromal cells with the ability to differentiate into other cell lineages such as cartilage, bone, and adipose tissues. Tissue-resident mesenchymal stem cells may have a role in repair, fibrosis, cardiovascular diseases, and tumor microenvironment. Studies have suggested that formation of myofibroblasts is associated with diseases such as cancer, fibrosis, and vessel disease. The Human Prostate Stromal Mesenchymal Stem Cells (HPS-19I) are isolated CD44+CD90+ cells from the ***** prostate gland which has the ability to differentiate into neurogenic, chondrogenic, and osteogenic cell lineages. In the presence of TGF-β1 the cells differentiate into myofibroblasts by inhibiting RUNX1 expression. The cells potentially are a source of reactive stroma fibroblasts. These cells are recommended in use for stem cell biology, stroma microenvironment, anticancer therapeutic and antifibrotic therapeutic studies.

Propagation Use of PriCoat™ T25 Flasks (G299) or . Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow III medium available from ABM (TM003). To make the completed growth medium, add the following components to the base medium to a final concentration of: 5% fetal bovine serum (TM999), 5% NuSerum, 0.5 µg/ml testosterone, 5 µg/ml insulin, and Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.

Note: Density-dependent inhibition is exhibited at more than 70% confluency.
