人角膜是一个独特的组织,因其透明且无血管。角膜包含三个不同的细胞层,外层上皮细胞、中间层基质和内层内皮细胞。角膜上皮细胞在先天免疫反应中起着作用,通过感应病原体存在并发出信号从而激活角膜防御系统。角膜上皮细胞还能高水平的表达乙醛脱氢酶,防止紫外线和4-羟基壬烯醛对细胞造成的伤害。角膜内皮细胞的更新由细胞因子调控,比如表皮生长因子,它能激活上皮细胞深层的同源受体。体外培养角膜内皮细胞已经可以替代毒理学检测中的动物实验。它们也可以被用来研究角膜细胞的增殖、分化和死亡之间的高度集成和平衡。
Sciencell研究实验室的HCEpiC是从人角膜分离得到的,原代冻存,冰冻运输。每管细胞密度*过5×10^5/ml。细胞特性经无血清培养基中镜检为鹅卵石形态,CK-18,和-19免疫荧光鉴定。本细胞经检测不含HIV-1、HBV、HCV、支原体、细菌、酵母菌和真菌。如采用ScienCell 实验室特制的培养基,可保证此细胞10倍增增殖。
