呼吸道上皮细胞是由纤毛、无纤毛和能分泌黏液的细胞按照从近端到远端呼吸道分布的混合细胞群组成。这类细胞的各个亚型细胞各具特点,从而不仅能形成物理屏障,可有效的防止多种有毒物质,而且通过产生和分泌大量化学介质和细胞因子形成高度复杂的宿主防御系统。支气管上皮由表面的上皮细胞和粘液腺体组成。表面的上皮细胞由基细胞,杯状细胞和纤毛细胞三种细胞构成,形成了基底柱状结构,并能清除粘液纤毛。用支气管上皮细胞培养分化诱导纤毛表型的研究显示,IL-4和IL-13的刺激能改变增殖、纤毛运动、黏液分泌等一系列细胞功能。支气管上皮细胞增殖以及其它细胞过程也部分受到EGF受体信号的调节。支气管上皮细胞的培养为我们研究这类细胞的功能和病理生理机制提供了一个体外模型。
人支气管上皮细胞提取于人支气管,原代冻存。每管含有细胞数>5×105 cells/ml,此细胞通过Cytokeratin-18, -19和 Vimentin抗体荧光免疫方法验证,经测试不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌。
